Assays

Overview

Once you have created your project, add the assays you are measuring.

Adding assays

The assays you measure or plan to measure for this project are based on the experimental objectives you aim to optimize or track on the Cradle platform. You can define as many assays as needed by following these steps:

  • Set an Assay name, a name that your team uses for a specific assay.

  • Choose a Task that refers to the type of protein characteristic you are measuring, such as activity, stability, or expression.

  • Select a Property to define the specific metric used to measure the Task. For example, for activity, this could be the Michaelis constant (Km) or the catalytic rate constant (kcat).

  • Set an Assay unit that you use to measure the specified Property.

Note: You can create a custom option if your Task, Property, or Assay unit is not listed in the dropdown menu.

Example

You are working on an antibody. The thing you care most about is the binding affinity. You have two assays at different temperatures to measure this.

In the Cradle platform, you create an assay and set Binding Affinity 45 °C as assay name.

You set the task as Binding and the property to Dissociation Constant (Kd). For assay unit you select μM (microMolar).

You repeat the same process for your other assay, and voila! You have now added your assays.
